Yes, if the code is weak spark or a coil that has broken down. On four-cylinder engines the cylinder codes run P0301 through P0304. On the 3.0L V6 they can run through P0306. P0300 means the misfire is moving between cylinders. Coils and iridium plugs fix the spark side, not a vacuum leak.
Why are Escape coils sold with spark plugs?

How hard is an Escape coil replacement?
